Biosynthesis and characterization of copolymer P(3HB-co-4HB) by Cupriavidus sp. using substrate mixtures containing 1,8-octanediol (PhD)

Copolymer P(3HB-co-4HB) is one of the polyhydroxyalkanoates (PHAs) which has received considerable attention as a potential biomaterial in medical and pharmaceutical applications. It was previously discovered that a high molecular weight P(3HB-co-4HB) was produced by wild type Cupriavidus sp. upon cultivation with substrate mixtures containing 1,8-octanediol. In this study, research will focus on developing fermentation strategies which would retain the copolymer properties in large-scale fermentation process (20 and 100 L). The properties of the copolymers produced will be characterized by using tensile machine, differential scanning calorimetry (DSC), gel permeation chromatography (GPC) and nuclear magnetic resonance (NMR).

Biosynthesis and characterization of copolymer P(3HB-co-4HB) by transformant Cupriavidus sp. using substrate mixture of 1,4-butanediol and 1,6-hexanediol (MSc)

PHA synthase is a polymerizing enzyme which plays a vital role in overall polyhydroxyalkanoates (PHAs) production. Transformant Cupriavidus sp. was constructed by adding extra phaC gene (responsible for PHA synthase) from Cupriavidus sp. This successfully constructed transformant was found able to produce P(3HB-co-4HB) with high 4HB monomer composition and PHA content. In the subsequent research, transformant strain will be employed to produce this P(3HB-co-4HB) copolymer in large scale fermentation (20 and 100 L). This project will screen and identify the best scale up parameters for the copolymer production. The properties of the copolymers produced will be characterized by using tensile machine, differential scanning calorimetry (DSC), gel permeation chromatography (GPC) and nuclear magnetic resonance (NMR).

Production of PHA using C1 compounds as the carbon source (PhD)

Industrial application of PHAs is largely hindered by the prohibitive cost of the substrates used, which can account up to 50% of the production cost. In this research, studies will be conducted using locally-isolated microorganisms to utilize methane or methanol as the production substrate. Optimization of the fermentation and polymer characteristics will also be conducted.

Optimization of the steam explosion and enzymatic hydrolysis for sugars production from palm oil empty fruit bunch (MSc)

Utilization of biomass for biofuel production has gained impetus in recent years as a means of utilizing renewable resources for energy production and also to mitigate the production of green house gases. The saccharification of cellulosics to produce sugars is the primary step in the utilization of cellulosics. In this project, a screening programme will be carried out to isolate high-yielding cellulolytic microorganisms effective in breaking down palm oil empty fruit bunch. The production of cellulases will be optimized and characterized.
